Аннотация

The fitness of male guppies (Poecilia reticulata) highly depends on the size and number of their black, blue, and orange ornaments. Recently, progress has been made regarding the genetic mechanisms underlying male guppy pigment pattern formation, but we still know little about the pigment cell organization within these ornaments. Here, we investigate the pigment cell distribution within the black, blue, and orange trunk spots and selected fin color patterns of guppy males from three genetically divergent strains using transmission electron microscopy. We identified three types of pigment cells and found that at least two of these contribute to each color trait. Further, two pigment cell layers, one in the dermis and the other in the hypodermis, contribute to each trunk spot. The pigment cell organization within the black and orange trunk spots was similar between strains. The presence of iridophores in each of the investigated color traits is consistent with a key role for this pigment cell type in guppy color pattern formation.

The rich variation in adult color patterns of male guppies (Poecilia reticulata) has attracted the attention of geneticists and ecologists for almost a century. Studies on their embryogenesis, however, have been limited by the fact that guppies are live bearers. We have observed normal development after explantation of guppy embryos from the ovary of pregnant females at various times after last parturition, and found that development of each batch of eggs is slightly asynchronous, most likely due to asynchronous fertilization.
We have cultured explanted embryos in vitro and continuously observed their development. Although embryos explanted a few days after fertilization survived up to 4 weeks in culture, they did not complete their development. In contrast, embryos explanted at late stages of gestation could hatch and develop to fertile adults. Our embryo culture techniques overcome some of the limitations of using livebearers as study objects, and they allow continuous observation of and accessibility to live embryos at all stages.

All too often in evolutionary biology we are led to speculate or infer the mode of action of natural selection; we usually do not know why some individuals are more adaptive than others. Very often attempts to measure natural selection are unsuccessful, leading to heated arguments abouf the relative importance of selection, genetic drift, and epistasis in evolution (Lewontin, 1974). Until we know more about how and why natural selection occurs, attempts to measure it are quixotic, and discussions of its importance are theandric.
It is no coincidence that most of the successful studies of natural selection have dealt with animal color patterns; it should be obvious which color patterns are more adaptive in the presence of visually hunting predators. The adaptive significance of warning coloration and mimicry of distasteful species has been worked out (Cott, 1940; Wickler, 1968; Edmunds, 1974; Rothschild, 197 5; Turner, 1977). But most species are neither distasteful nor mimetic; most have inconspicuous or cryptic color patterns in their natural habitats (Poulton, 1890; Thayer, 1909; Cott, 1940; Endler, 1978).
Most field and experimental studies have shown that the overall color or tone of inconspicuous species matches or approximates the background (DiCesnola, 1904; Sumner, 1934, 1935; Isley, 1938; Popham, 1942; Dice, 1947; Kettlewell, 1956, 1973; Turner, 1961; Kaufman, 1974; Wicklund, 1975; Curio, 1976), but they treated species with solid colors or have ignored color pattern. A color pattern can be regarded as a mosaic of colored spots or patches of various sizes, colors and shapes. Some striking examples of color pattern polymorphisms remain largely unexplained, for example in Partula and Cepaea (Clarke and Murray, 1971; Jones et al., 1977). In three fish species, the frequency of a color pattern element is correlated with the presence or absence of visually directed predation: Gasterosteus aculeatus (McPhail, 1969; Semler, 1971; Moodie, 1972), Nothobranchius guntheri (Haas, 1976a, b), and Cichlasoma citrinellum (Barlow, 1976; Barlow and Ballin, 1976; McKaye and Barlow, 1976). But these simple pattern polymorphisms tell us very little about the factors which determine color patterns as a whole. The purpose of this paper is to show how various aspects of color patterns are moulded by natural selection.

The food and feeding habits of the Guppy, Poecilia reticulata, from drainage canal systems in Lagos Metropolis, Nigeria, was investigated over a period of 24 months. Fish samples were collected monthly from 15 study sites. A total of 2400 fish stomachs were analyzed using the numerical and frequency of occurrence methods. P. reticulata fed mainly on algae, organic detritus, diatoms, mosquito larvae parts, protozoan, zooplankton and fish parts while algae form the most abundant and important food item, constituting 79.03% of food items by numerical and 33.17% by occurrence of stomachs examined. Amongst the algae, Ulothrix sp. was the most preferred, constituting 33.17% by numerical and 20.82% by occurrence. The least eaten food item was fish parts constituting 4.46% by numerical and 1.19% by occurrence. There was a significant difference (P < 0.05) in the number of algae and mosquito larvae consumed, while there was no significant difference (P > 0.05) in the number of other food items consumed for both seasons. However, by occurrence method there was no significant difference (P > 0.05) in food items consumed for both seasons. The three size classes of P. reticulata exhibited similar food habits with the presence of eight categories of food items in their stomachs. The largest size class ate more of algae, organic detritus and fish parts, followed by the medium size class while the small size class ate less of these food items. The species is an opportunistic benthopelagic omnivores, whose preference for food fluctuates with season, with a peak in diversity of food types occurring in the rainy season.

Males of the guppy (Poecilia reticulata) vary tremendously in their ornamental patterns, which are thought to have evolved in response to a complex interplay between natural and sexual selection. Although the selection pressures acting on the color patterns of the guppy have been extensively studied, little is known about the genes that control their ontogeny. Over 50 years ago, two autosomal color loci, blue and golden, were described, both of which play a decisive role in the formation of the guppy color pattern. Orange pigmentation is absent in the skin of guppies with a lesion in blue, suggesting a defect in xanthophore development. In golden mutants, the development of the melanophore pattern during embryogenesis and after birth is affected. Here, we show that blue and golden correspond to guppy orthologs of colony-stimulating factor 1 receptor a (csf1ra; previously called fms) and kita. Most excitingly, we found that both genes are required for the development of the black ornaments of guppy males, which in the case of csf1ra might be mediated by xanthophore–melanophore interactions. Furthermore, we provide evidence that two temporally and genetically distinct melanophore populations contribute to the adult camouflage pattern expressed in both sexes: one early appearing and kita-dependent and the other late-developing and kita-independent. The identification of csf1ra and kita mutants provides the first molecular insights into pigment pattern formation in this important model species for ecological and evolutionary genetics.

Background: Next-generation sequencing is providing researchers with a relatively fast and affordable option for
developing genomic resources for organisms that are not among the traditional genetic models. Here we present a de novo assembly of the guppy (Poecilia reticulata) transcriptome using 454 sequence reads, and we evaluate potential uses of this transcriptome, including detection of sex-specific transcripts and deployment as a reference for gene expression analysis in guppies and a related species. Guppies have been model organisms in ecology, evolutionary biology, and animal behaviour for over 100 years. An annotated transcriptome and other genomic tools will facilitate understanding the genetic and molecular bases of adaptation and variation in a vertebrate species with a uniquely well known natural history.
Results: We generated approximately 336 Mbp of mRNA sequence data from male brain, male body, female brain, and female body. The resulting 1,162,670 reads assembled into 54,921 contigs, creating a reference transcriptome for the guppy with an average read depth of 28×. We annotated nearly 40% of this reference transcriptome by searching protein and gene ontology databases. Using this annotated transcriptome database, we identified candidate genes of interest to the guppy research community, putative single nucleotide polymorphisms (SNPs), and male-specific expressed genes. We also showed that our reference transcriptome can be used for RNA-sequencing-based analysis of differential gene expression. We identified transcripts that, in juveniles, are regulated differently in the presence and absence of an important predator, Rivulus hartii, including two genes implicated in stress response. For each sample in the RNA-seq study, >50% of high-quality reads mapped to unique sequences in the reference database with high confidence. In addition, we evaluated the use of the guppy reference transcriptome for gene expression analyses in a congeneric species, the sailfin molly (Poecilia latipinna). Over 40% of reads from the sailfin molly sample aligned to the guppy transcriptome.
Conclusions: We show that next-generation sequencing provided a reliable and broad reference transcriptome.
This resource allowed us to identify candidate gene variants, SNPs in coding regions, and sex-specific gene expression, and permitted quantitative analysis of differential gene expression.

The effects of feeding guppy (Poecilia reticulata) fry a diet offered as either powder or £akes on their growth and survival were tested over a period of 8 weeks. The results show that the growth of both male and female fish was considerably enhanced when the diet was
presented in the form of a finely ground powder compared with a £ake form. Final average weights of fish given a diet containing the exact same ingredients (44.9% protein and 6.1% fat), from the same
batch of raw materials in the powdered form were 280.0±12.1mg compared with 114.6±19.9mg for the diet given in the form of flakes. In a diet that had a higher fat level (45.1% protein and 10.6% fat), the difference in final weight attained was even more
dramatic: 303.9±16.7mg for the powder-fed fish
compared with 92.6±12.5mg for the flake-fed fish.
The coefficient of variance in the fish fed a £ake diet was signi¢cantly higher than that exhibited in the treatments fed the powder feed. The ability of the ¢sh to consume the food in a rapid manner, preventing leaching of vital nutrients from the feed before being engulfed by the ¢sh probably led to the better
growth results exhibited by the fish given the powdered food.
